Since Harvard Professor David Sinclair discovered in 2013 that NMN could extend the lifespan of mice, the anti-aging effects of the "fountain of youth" NMN have been continuously explored.

Human clinical trials have confirmed several NMN anti-aging benefits, including improved insulin sensitivity, delayed arterial hardening, muscle atrophy, skin aging, and intestinal aging.

Recently, a team led by Professor Haruka Wada of Hokkaido University in Japan discovered that NMN also has anti-cancer effects.

This new study, published in the Journal for ImmunoTherapy of Cancer, demonstrates that cancer stem cells trigger macrophage aging (macrophages are the body's first line of defense against cancer cells), but supplementing NMN to mice inoculated with cancer stem cells can improve this condition, significantly preventing tumor growth, prolonging mouse survival time, and increasing their survival rate.

First, researchers co-cultured cancer stem cells with immune cells (including macrophages and T cells) in a medium to observe whether cancer stem cells would affect the activity of these immune cells.

Macrophages can kill pathogens, absorb apoptotic and necrotic cells, and play important roles in anti-infection and anti-tumor functions; T cells can more specifically identify and eliminate specific pathogens (avoiding the accumulation of pathogens leading to cancer), and "remain vigilant" even after recovery, providing long-term protection for the body.

The results showed that the proliferation of macrophages was significantly inhibited in terms of quantity. Not only that, these macrophages appeared flat and enlarged, showing a morphology closer to a "senescent state."

To confirm whether these "senescent" macrophages were indeed senescent, researchers used one of the most widely used biomarkers of senescence, SA-β-gal, to measure the degree of macrophage senescence: an increase in SA-β-gal activity is one of the main characteristics of cell senescence.

The results showed that the SA-β-gal activity in these macrophages had reached the standard for determining them as senescent cells.

Furthermore, in the subsequent analysis of "cell senescence-related genes," researchers also found that these macrophages highly expressed p21 and Glb1, two genes known to be closely related to cell senescence.

These findings all prove that macrophages co-cultured with cancer stem cells have indeed entered a senescent state.
